Flight MU745 departed Shanghai Pudong International Airport at 2:19 a.m. local time, the media reported.
The mammoth journey spans nearly 20,000 kilometers (12,427 miles) and necessitates a refueling stopover in Auckland, New Zealand.
The carrier asserts this route establishes a new benchmark as the world's longest one-way flight operation.
Passengers can expect travel time reductions exceeding four hours when journeying between China and South America, the airline announced.
China Eastern Airlines will maintain two round-trip services weekly along this newly established route.
